(Dis-)Connected Dots in Dementia with Lewy Bodies—A Systematic Review of Connectivity Studies

Dementia with Lewy bodies (DLB) is characterized by widespread connectivity disruptions compared to healthy elderly controls. Across the reviewed structural and functional neuroimaging modalities disruptions emerge most consistently in frontoparietal, default mode, and visual networks. Impairments in brain connectivity measures were associated with clinical features of DLB patients.

Proximity Extension Assay technology applied in longitudinal plasma samples from drug-naive patients with Parkinson's disease and matched healthy control patients from the de novo Parkinson's cohort demonstrated marker candidates for state, fate, and disease progression. Reduced endothelial/atherosclerotic processes, increased thromboembolic risk, and potentially reduced α-synuclein cleavage could be validated in an independent, multicentric Parkinson Progression Marker Initiative cohort.
